Instance should be in the custom VPC, and the corresponding subnet, security group; route table should be connected with the gateway, gateway has to be attached to the custom VPC.

To connect to an EC2 instance in a custom VPC: Public Internet Access: Instance in a public subnet (route to Internet Gateway). Instance has a public IP address (auto-assigned or Elastic IP). Security Group allows inbound traffic (SSH/RDP) from your IP. Subnet's route table directs internet traffic to the IGW. If instances is in Private VPC/Peered VPC Access: Instance can be in any subnet. Ensure network connectivity (local VPC routing or peering routes). Security Group allows inbound traffic (SSH/RDP) from the source's private IP range.
